The BCA Protein Assay Kit provides a simple, rapid, detergent tolerant (up to 5%) procedure for determining protein concentration. The BCA method takes advantage of the fact that copper can be reduced from the cupric to the cuprous state by the peptide bond of protein. Cysteine, histidine, tyrosine, tryptophan and methionine also contribute to the process. The cuprous ion forms a complex with 2 bicinchoninic acid molecules in basic solution with a very strong absorbance band at 562 nm. The intensity of the color is proportional to the amount of protein in a sample. The BCA Protein Assay is useful for protein amounts in the range of 0.5 – 30 µg (concentrations of 0.01 - 0.6 mg/ml).
