HDL and LDL play a role in various pathological conditions. Low levels of HDL combined with high level of LDL are associated with an increased risk of cardiovascular events including plaque rupture, stroke, hypertension, etc. AkrivisBio’s HDL/LDL Assay provides a simple, sensitive method for quantifying HDL and LDL/VLDL cholesterol separately. In this assay, cholesterol oxidase oxidizes free cholesterol with the formation of hydrogen peroxide. Hydrogen peroxide is then utilized by the peroxidase to oxidize ADHP thereby resulting in the formation of intense color or fluorescence measured at OD 570 nm or at Ex/Em = 538/587 nm respectively.
In an optional step, cholesterol esterase cleaves cholesterol esters to free cholesterol, permitting the assessment of free and total cholesterol separately. Thus, if cholesterol esterase is included in the assay, total cholesterol is measured. If it is not included, free cholesterol is measured. The amount of cholesterol ester can be calculated by subtracting free cholesterol from total cholesterol.
