Triglycerides are prime storage molecules. Lipolysis is the process by which they are converted to glycerol and free fatty acids, supplying the body with readily metabolizable sources of energy. Lipolysis occurs primarily in white and brown adipose tissue. Deficiencies in lipolysis lead to increased intracellular lipid accumulation, resulting in many of the hallmarks of metabolic syndrome - abnormal cellular physiology, hyperlipidemia, and insulin resistance. Lipolysis is induced by several triggering chemicals, among them catecholamines and inhibited by insulin. AkrivisBio’s Lipolysis Assay is a simple, sensitive means of measuring lipolysis in cultured cells such as 3T3 cells. Stimulating cells with isoproterenol starts the cascade resulting in release of free glycerol which is measured in the assay with an enzyme linked assay leading to color and fluorescence with a lower limit of detection of less than 1 micromolar.
