Elastin is a protein of the extracellular matrix that provides elasticity to tissues. It is primarily present in extensible tissues including arteries, lung, ligaments and skin to resume their shape after stretching or contracting. Due to its role in normal development and organ function, impairment of elastin synthesis or proteolytic degradation of elastin fibers results in major clinical pathologies. Mutations in the Elastin gene lead to diseases such as Williams-Beuren syndrome, Cutis laxa and Supravalvular aortic stenosis. Several skin diseases have been associated with abnormalities of elastin and marked fragmentation of elastin is observed in pulmonary emphysema. AkrivisBio’s Elastin Assay is a quantitative dye-binding method for the analysis of elastin in tissues and cultured cells. Due to its unique chemistry and highly cross-linked nature, elastin is stable. In this kit, elastin is denatured and solubilized under high heat and extreme pH conditions. Isolated elastin binds to a dye to form a colored product, measured at 513 nm. The signal is directly proportional to the elastin content. The assay is simple, reliable and selective with a linear range of 0 μg-40 μg elastin per well.
