Hexokinase catalyzes the first step of glycolysis, the phosphorylation of glucose. Through phosphorylation, glucose becomes a negatively charged molecule, unable to diffuse across the cell membrane unlike its glucose predecessor, trapping it within the cell. There are four isoforms of hexokinase. Type IV, also called glucokinase, has a low affinity for glucose and is used metabolically to help regulate blood sugar concentrations. Hexokinase dysregulation is associated with diseases such as X-linked muscular dystrophy and an autosomal recessive hemolytic anemia as well as the important pharmacological targets, diabetes and cancer. Since cancer cells frequently exhibit increased glycolysis, hexokinase has become a key target of research.
