Purine Nucleoside Phosphorylase is involved in the purine salvage pathway, recycling purines for nucleotide synthesis. It catalyzes the phosphorolysis of inosine and guanosine, breaking them down into their respective bases, hypoxanthine and guanine and ribose-1-phosphate. PNP deficiency leads to a rare immunodeficiency disorder with a severe effect upon T-cells while often not affecting B-cells. In the disease, dGTP accumulates in the T-cells, inhibiting ribonucleotide reductase and DNA synthesis, leading to dysfunction and apoptosis. Altered purine metabolism has been observed in some cancers and PNP is a potential target as a therapeutic strategy.
