The ability to quantify the number of viable cells in a system under controlled conditions is a requirement in cell biology research. The classic cell proliferation assays all measure the metabolic activity associated with cell samples. The assumption is that conditions preventing cell proliferation are reflected in the lack of increased metabolic activity. This is normally true except under conditions of cytostatic agents which can prevent cell division but allow for cell growth without division. In this case, increases in cell size along with increases in the numbers of subcellular organelles such as mitochondria can disguise the effect the cytostatic agents are having on the cells. The solution is to monitor the amount of DNA in a sample. Since DNA duplication associated with cell division is suppressed, the effects of drugs can be observed under conditions where classical cell proliferation assays such as MTT and MTS will miss the effect. AkrivisBio’s DID cell proliferation assay uses HO33342 to quantitate the total amount of DNA in a well. Its cost-effectiveness, versatility (adherent or suspension cultures, 96 or 384 well plates) and the ability to ignore metabolic activity makes it well adapted for cell proliferation assays.
