Glutathione is the major intracellular low-molecular-weight thiol in biological systems. It works against oxidative stress in tissues and cells. Most glutathione assays, such as the DTNB-enzyme cycling glutathione assay or the Monochlorobimane based assay don’t distinguish between reduced and oxidized glutathione. The AkrivisBio Glutathione Assay provides detection of GSH, GSSG, and total glutathione separately. In the assay, GSH reacts but not GSSG, generating fluorescence, allowing specific quantitation of GSH. Subsequently, addition of a reducing agent converts GSSG to GSH, permitting determination of total (GSH + GSSG). In order to measure GSSG specifically, a GSH Quencher is added, removing GSH, while GSSG is unaffected. Addition of a reducing agent converts GSSG to GSH which is measured as before, thus allowing for specific GSSG quantitation. We provide a buffer which eliminates protein thiol interference and stabilizes GSH in solution. Sensitivity is ~2-400 ng/ml of glutathione.
