Enolase plays a central role in glycolysis. It converts 2-phosphoglycerate to phosphoenolpyruvate. It also catalyzes the reverse reaction (PEP to 2-phosphoglycerate) during gluconeogenesis. Enolase has been found in every organism with a glycolytic pathway. Other than its role in glycolysis, enolase has been found to affect other functions such as transcriptional regulation, cell adhesion and stressor response. Extracellular enolase interacts with the extracellular matrix and proteins like laminin and fibronectin. Cancer cells may upregulate enolase to enhance their adhesion to the ECM, facilitating metastasis.
