ECE-1 assists in the regulation of vascular tone and blood pressure through its involvement in the endothelin system. ECE-1 is a membrane-integrated metalloproteinase with an extracellular zinc-binding active site. It is responsible for the final step of converting big endothelins into the biologically active forms. ECE-1 activity affects several physiological processes, namely vasoconstriction, cell proliferation, inflammation and fibrosis. It is widely expressed in human tissues with notably high levels in vascular endothelial cells. Dysregulation of ECE-1 is implicated in several pathological conditions- hypertension and cardiac disease, chronic kidney failure, cancer and Alzheimer's disease. The possibility of impacting these disease states makes it an attractive target for drug development.
