Enteropeptidase, also known as Enterokinase, is a member of the serine protease family, localized to the brush border of the duodenal and jejunal mucosa. Its primary function is to initiate intestinal digestion by activating trypsinogen to its active form, trypsin. This initiates a cascade of enzymatic reactions, activating other proenzymes to their active forms including chymotrypsin, elastase, carboxypeptidase and lipase. Enteropeptidase has a very specific substrate recognition motif of Asp-Asp-Asp-Asp-Lys (DDDDK). Due to its broad involvement in digestion, it has become a target of interest for pharmaceutical companies.
