Fumarase, with separate forms found in mitochondria and the cytosol, catalyzes the reversible hydration of fumarate to L-malate, playing a role in several metabolic pathways. The mitochondrial form is an essential component of the TCA cycle. The cytosolic form can translocate to the nucleus where it is involved in the DNA Damage Response participating in the non-homologous end joining repair pathway. Fumarate formed inhibits lysine demethylase 2B, facilitating histone H3 methylation and promoting double-strand break repair. Fumarase has been identified as a tumor repressor and its deficiency is linked to development of cancers. The presence of fumarase is conserved across species, suggesting an ancient origin for its role in genome stability.
