Glycerol-3-Phosphate Dehydrogenase (GPDH) links carbohydrate and lipid metabolism. There are two forms, cytosolic and mitochondrial, which work in concert to maintain metabolic balance and energy production. Cytosolic GPDH, located on the outer mitochondrial membrane, catalyzes the reduction of dihydroxyacetone phosphate to glycerol-3-phosphate, while the mitochondrial GPDH, embedded in the inner mitochondrial membrane, oxidizes G3P back to DHAP, enabling a “shuttle” of G3P across the membrane, important for the NAD/NADH balance in the cell. GPDH influences insulin secretion, lipogenesis in adipocytes, and both lipid and carbohydrate metabolism in the liver, exemplifying its importance in multiple ways.
