The conversion of isocitrate to α-ketoglutarate is a major regulatory step of the TCA cycle and has profound implications for energy metabolism, redox balance and many biosynthetic pathways. NADH or NADPH are both formed by different isoforms of isocitrate dehydrogenase. While NADH is important for energy metabolism, NADPH is important for redox balance, keeping glutathione in the reduced state. Isocitrate provides a carbon skeleton for nitrogen assimilation in plants, supports the glyoxalate cycle and has a role in glucose sensing and insulin secretion in pancreatic β cells.
