Formaldehyde (HCHO) serves as an intermediate in one-carbon metabolism. It is rapidly condensed with tetrahydrofolate to enter the 1C cycle, providing essential units for nucleotide biosynthesis, particularly important when serine catabolism is impaired. Formaldehyde's dual nature as both a potential toxin and a valuable metabolic resource is largely maintained by alcohol dehydrogenase 5, which not only detoxifies HCHO but also supports essential metabolism. An alternative pathway involves the reaction of HCHO with cysteine and histidine, forming timonacic and spinacine which can act as a HCHO buffer reservoir. HCHO affects glucose metabolism and impacts cellular energy metabolism.
