Histone deacetylase 6 (HDAC6) is a unique member of the HDAC family, primarily localized in the cytoplasm, that plays a significant role in various metabolic processes. Its importance is due to its ability to regulate multiple cellular functions. While normally associated with ϵ-amino deacetylation of histones, it has other important functions as well. HDAC6 is an essential regulator of hepatic glucocorticoid-stimulated gluconeogenesis. HDAC6 deficiency impairs dexamethasone-induced hepatic glucocorticoid receptor translocation, leading to attenuated expression of gluconeogenic genes. HDAC6 inhibitors, such as tubastatin A, have demonstrated potent anti-obesity effects in diet-induced obese mice. These inhibitors reduce food intake, fat mass, and hepatic steatosis while improving systemic glucose homeostasis. It is also involved in lipid metabolism, cellular stress response and cytoskeletal regulation through α-tubulin modification.
