Transketolase sits at the intersection of the pentose phosphate pathway and glycolysis and participates in the Calvin Cycle. It reversibly transfers a 2-carbon fragment from xylulose-5-P onto either ribose-5-P forming sedoheptulose-7-P and glyceraldehyde-3-P or to erythrose-4-P forming fructose-6-P and glyceraldehyde-3-P. This process is a significant source of NADPH and is important for maintaining the redox state of the cell. The reverse reaction is crucial for formation of ribose-5-phosphate. Transketolase pays a significant role in the Warburg effect and is associated with increased metabolism in cancer, where inhibition promotes tumor cell apoptosis. It appears to play a role in lipid metabolism and in neurological health.
