A TIBC / Serum Iron assay measures the % saturation of transferrin in serum. In the assay, an iron solution is added to one of two serum samples. Excess iron is removed, and the remaining iron is measured. The difference between the two denotes the amount of available iron binding sites there are in the sample. One of two more samples is treated to release any iron bound to transferrin then the two samples are measured. The difference is the amount of iron bound to transferrin. The ratio of iron bound to transferrin/the total available binding sites gives the % of transferrin sites binding iron or % saturation.
